基督教聖經之智慧書導讀 :	Dipyridamole在臨床上已經使用了很長一段時間，但是在近年來仍有研究將dipyridamole應用在其他適應症上，像是COVID-19或者其他特殊的心血管疾病。但是因為藥物在鹼性環境下溶解度不好，導致藥物的生體可用率不佳。而胃滯留劑型在過去的研究中有用來改善在鹼性環境下溶解度不好的藥物，所以本研究透過將Kollidon SR以及PEO 7000K以不同比例混合後填充膠囊，使用低複雜度的步驟將dipyridamole製備成胃滯留膠囊。將Kollidon SR加入至劑型中，可以加強劑型的抗溶蝕能力，溶蝕比例從71.9%下降至38.6%，而且對於溶離以及膨脹能力的影響不大，所以可以透過加入Kollidon SR來加強劑型的胃滯留效果。PEO 7000K的變化處除了會影響膨脹及溶離以外，對於長時間的溶蝕抵抗能力也會造成影響。PEO 7000增減在12h的斜率絕對值為171.46比起Kollidon SR增減在12h斜率絕對值為46.259高出許多，代表PEO 7000K對於抗溶蝕的影響的程度比Kollidon SR還要劇烈。除了PK-0:1以及PK-0.25:2以外，大部分處方都沒有漂浮的延遲時間而且漂浮時間都可以維持到12小時。就實驗結果來看，PK-0.33:2是較為理想的處方，因為在藥動試驗中與市售品比較的相對生體可用率 (268.2089.33%)在各個處方中為最高，而且也具有不錯的膨脹能力及抵抗溶蝕的能力，同時也具有理想的漂浮時間。就實驗結果看來，雖然主要是PEO 7000K在影響膨脹，抗溶蝕以及溶離速率，但是Kollidon SR 也可以達到增強劑型抗溶蝕能力，同時不會影響劑型的溶離和膨脹能力。可以透過先找到理想的PEO 7000K比例達到預期的溶離以及膨脹效果，在透過Kollidon SR 來補足抗溶蝕能力。.

520 3 $a Dipyridamole has been used clinically for a long time, but in recent years, there has been many research to explore its application in other indications. However, its poor solubility in alkaline environments has led to poor bioavailability.Therefore, this study aimed to prepare a gastroretentive capsule of dipyridamole by combining Kollidon SR and PEO 7000K in different ratio through a simple process. Adding Kollidon SR into the formulation enhanced the formulation's resistance to erosion, reducing the erosion from 71.9% to 38.6%, with little effect on dissolution and swelling. On the other hand, PEO 7000K not only affected swelling and dissolution but also affected the long-term erosion. PEO 7000 showed a higher slope (171.46) at 12 hours compared to Kollidon SR(46.259), indicating that PEO 7000K had a more effect on erosion than Kollidon SR. Except PK-0:1 and PK-0.25:2 formulations, most of formulations did not have floating lag time, and their floating time could be maintained for up to 12 hours. Based on the results, the PK-0.33:2 is more optimal. This is because of its relative bioavailability in pharmacokinetic tests compared to commercially products (268.20 ± 89.33%) is the highest among all formulations. Additionally, it exhibits favorable swelling and erosion resistance capabilities, along with an ideal floating duration. While PEO 7000K primarily affected swelling, erosion, and dissolution rate, Kollidon SR also enhanced the formulation's erosion resistance without decreasing dissolution and swelling abilities. The ideal amount of PEO 7000K can be determined to achieve the desired dissolution and swelling effects, with Kollidon SR enhancing the erosion resistance..
