The emergence of viral outbreaks has posed a great human threat and severely affected communities, public health infrastructures, and economies globally. In the last 20 years, the world has been hit by several and significant viral diseases, including the 2003 outbreak of severe acute respiratory syndrome coronavirus (SARS-CoV), the 2009 outbreak of H1N1 influenza virus, the 2012 outbreak of Middle East respiratory syndrome coronavirus (MERS-CoV), the 2015 outbreak of Zika virus (ZIKV), the 2020 outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and the 2022 outbreak of monkeypox virus (MPOX). These outbreaks caused severe damage overall to the countries affected but more importantly, they also demonstrated how unprepared the world is for these emerging pathogens. Despite efforts, the prophylactics and therapeutics developed to combat these viruses were not effective and/or long lasting, urging to identify better approaches. In this dissertation, the goal was to utilize drug repurposing, e.g. drugs already approved or studied for the treatment of a new disease, as an approach to identify broad-spectrum antivirals that could be used against emerging viral infections. This is possible due to the target of the compound being a host factor that is required by different viruses to replicate. While identifying therapeutics against ZIKV, SARS-CoV-2 emerged so we applied our expertise in drug discovery to identify therapeutics against SARS-CoV-2 . Utilizing compounds from ReFRAME and NPC libraries, we were able to identify inhibitors against ZIKV and SARS-CoV-2.
